Kavain and dihydrokavain are presumed to be the most permeable to the blood–brain barrier. Kavalactones facilitate the functioning of GABA A receptors, in a manner that is similar to benzodiazepines, but kavalactones do not appear to bind to benzodiazepine receptors. The overall effect involves positive allosteric modulation of chloride channels, increasing the influx of chloride and hyperpolarizing the cell.
